Fluorometry

NEONATAL PKU Screening Fluo

The NEONATAL PKU Screening Fluo is an enzymatic test designed to screen newborns affected by phenylketonuria. This device allows the fluorescence determination of the quantity of phenylalanine presents in a drop of blood dried on 903® or 226 blotting paper.

Parameter screened

  • Phenylalanine (Phe)

Test Principle

Phenylalanine, extracted from neonatal samples, is combined with the enzymatic reagent phenylalanine dehydrogenase. In the presence of NAD+, the enzyme catalyzes the oxidative deamination of phenylalanine to phenylpyruvate and generates NADH. NADH acts as a co-factor for the reduction of resazurin to resorufin by the enzyme diaphorase. The intensity of the resorufin fluorescent signal (emission at 590 nm after excitation at 544 nm) is directly proportional to the concentration of Phenylalanine present in the sample.

Product Benefits

Easy-to-use : No transfer step required

Needs little biological material: use of only one punch

Practical: Identical protocol to the " NEONATAL MSUD Screening Fluo " and " NEONATAL TGAL Screening Fluo " devices

Fast: Results available in only 1h15

Sensitive: Increased sensitivity of the test due to the use of the fluorescence technique

Certification: Under notified body revision to comply with the IVDR standard

Illness

This test is designed to screen newborns for phenylketonuria using a blood sample dried on 903® or 226 blotting paper.
